Biological samples are inhomogeneous in nature due to complex processes taking place at cellular and sub-cellular levels. During disease progression, different types of changes, in structure, function, or composition, happen at both these levels. Our aim is to discern these changes using non-invasive methods involving light-tissue interactions. There are multiple events such as absorption, scattering and reflection off the surface, during interaction of light with a biological sample. Laser Speckle Imaging (LSI) is a modality based on the scattering phenomenon of light and gives a cumulative effect of change in optical scattering of the media. LSI has been extensively used to capture the changes in functionality, such as assessing the microcirculatory flow and finds application in the field of ophthalmology, neuroscience, and dermatology. Laser speckle parameters for extracting structural differences such as spatial disorganization and orientation changes of cellular structures has been explored here. Preliminary studies have been performed on analysing structural differences, both at gross and fine level, in tissues utilizing phantoms, bulk tissue specimens and on tissue sections. The proposal also aims to extend the study onto other biological samples such as biofilms and utilizing speckle parameters to extract structural changes in growth of biofilms.
